New methodology for genomic characterization of S-alleles in Japanese plum cultivars

| dc.description.abstract | Prunus species have a gametophytic incompatibility system that prevents self-fertilization and promotes crosspollination. This system is regulated by the multiallelic S locus, formed by two closely linked genes, the S-RNase expressed in the pistil and the SFB in the pollen tube. The identification of both S alleles of a cultivar is essential to establish its pollination needs, as well as its suitability as a parent in breeding programs. To date, in Japanese plum (hybrids of Prunus salicina Lindl.), 57 S alleles have been described (42 S-RNase and 15 SFB), which has allowed the assignment of more than 200 cultivars to 26 incompatibility groups. However, identification of S alleles has not been possible in a number of cultivars, presumably because their S alleles lack length polymorphism with already known alleles and have not been previously sequenced. In this work, we resequenced the genome of nine Japanese plum cultivars and applied a novel NGS-based methodology for S-genotyping and targeted de novo assembly of S-alleles and loci. | |
